I use Photoshop CS5 and a Wacom Bamboo Touch tablet (slowly dying)...
I use TONS of layers sometimes to get my shading done.

With lineless drawings such as my running Reno I start off with a sketch, then refine it to what I want to paint over. On another layer I begin blocking in the base color (usually one solid color just to cover the area) then lock opacity so I can't color outside my object and add any markings. Then I use multiple other layers to get the shading done.

With the foal tag, I pretty much just scanned my sketch into my computer and drew/colored over it. I think I have about four or five layers of color and shading on the base .psd file (I make recolors of certain things, such as that tag).
