Only one absolute way to know, have her preg checked by the Vet.

You can rig a catch if your mare is compliant. Otherwise figuring out their schedule and being close at hand around pee time can get your needed sample. Typically they pee every 6 hours or so. I have those that go before feeding in anticipation, one that pees while she eats - never fail, and a few that pee within a half hour after...
